humancolorectalcancer
Objective：ToresearchtheexpressionofmiR-196bandHoxB8mRNAincolorectal
cancerandtheircorrelationwithclinicopathologicfeatures,toexploretherelationship
betweenmiR-196bandHoxB8invivo.
Methods：ExpressionofRNA(includingmiR-196bandHoxB8mRNA)andHoxB8
proteinwere detectedrespectivelyby quantitativereal-timereversetranscriptasePCR
and western blot in 30 cases of colorectal carcinoma and corresponding normal
mucousmembranetissue.
Results：In colorectal carcinoma, expression of miR-196b and HoxB8mRNA were
higherthanthatofthecontrol(P＜0.05).ExpressionofmiR196bwasassociatedwith
lymphnodemetastasis，neoplasmstages(I+II,III+IV)anddistantmetastasis(P＜0.05).
Ontheotherhand,nosignificantdifferenceswereobservedregardingtumorsite,size,
grosstype,depthofinvasion,tissuedifferentiation,ageandsex(P＞0.05).Expression
of HoxB8mRNA was no significant differences concerning lymph node metastasis,
tumor stages(I+II,III+IV), distant metastasis, tumor site, size, gross type, depth of
invasion,tissuedifferentiation,ageandsex(P＞0.05).Incolorectalcancer,expression
of HoxB8 protein was lower than that of the control(P＜0.001). The expression of
miR-196b mRNA was negatively correlated with HoxB8
mRNA expression
（r=-0.458，P＜0.05), andHoxB8protein expressionwithno obviouscorrelation
incolorectalcancer （r=-0.236，P＞0.05).
Conclusions：(1) The expressions of miR-196b and HoxB8 mRNA in colorectal
cancer are higher,
the high expression of miR-196b mRNA is related to the
tumorigenesis and progression of colorectal cancer as well as correlated with
prognosis in colorectal cancer. (2) The miR-196b inhibits expression of HoxB8
mRNAbybindingtothe3′-UTRoftargetHoxB8mRNA.
