1.将托盘中的水加至快满为止。 2.制备滤纸片。将大小相同的 8 片滤纸片放在培养皿里的鲜肝匀浆中(含过氧化氢酶)浸 泡 1min,然后用镊子夹起滤纸片,靠在培养皿壁上,使多余的匀浆流尽。 3.准备反应小室。
①将 2 片含有酶的滤纸片小心放入反应小室的一侧内壁上, 使滤纸片粘在内壁上, 提醒学生 注意滤纸片不要碰到反应小室的瓶口。 ②将反应小室稍立起,使贴有滤纸片的一侧在上面,小心加入 pH 5.0 的缓冲液 2mL,然后 再加入 2mL 30%的 H2O2 溶液,切勿使上述混合液接触到贴在内壁上的滤纸片。并将反应 小室的瓶口塞紧。 4.收集 pH5.0 的条件下,反应小室中产生的气体量。 ①将 25mL 量筒横放于盘中使之灌满水,若有气泡,将其轻轻倾斜,小心赶出气泡。将量筒 倒立,使筒口一直处于水中。 ②小心将反应小室平放在盘中的水里, 注意反应小室贴有滤纸片的内壁应在上面, 将量筒移 至反应小室口上伸出的玻璃管上方,实验过程中要一直扶着量筒,保证量筒的位置不动。 ③将反应小室小心旋转 180°,使 H2O2 溶液接触滤纸片。同时开始计时,分别在 0.5min 和 1min 时,读取量筒中水平面的刻度并作好标记,然后记录。
